Full Description

The U.S. Environmental Protection Agency's (USEPA's) National Homeland Security Research Center (NHSRC) is currently working with laboratories and subject matter experts (SMEs) to optimize and verifyculture-based methods for S. Typhi and V. cholerae O1 and O139 in environmental matrices (e.g., drinking water,surface water). Once the culture component has been optimized, a polymerase chain reaction (PCR) protocol willbe optimized for use with the enrichment step included in each culture method; this will allow laboratories to utilizethe same upfront processing for either culture or molecular analyses. Most of the available culture-based protocolswere not developed or optimized for environmental matrices, requiring some method development work to addressbackground flora, complex environmental matrices (e.g., surface waters), and spiking procedures to evaluaterecoveries.
